Leuprolide

WHAT IS IT?

Leuprolide (Lupron) is a GnRH analog that has agonist properties with used in a pulsatile fashion (increases FSH/LH), and antagonistic properties when used continuously. When used continuously it down regulates GnRH receptors in the pituitary, leading to decreased production/release of both FSH/LH. For more on the gonadal hormone axis look here.

WHEN DO WE USE IT? 

Pulsatile: Infertility

Continuous: Prostate cancer (adenocarcinoma), uterine fibroids, precocious puberty

WHEN DO WE AVOID USING IT?

Known/suspected pregnancy, breast feeding

HOW IS IT ADMINISTERED?

Intramuscular or sub-cutatneous injection

WHAT ARE THE SIDE EFFECTS/TOXICITY?

Anti-androgen effects, nausea, vomiting

Suggested risk of diabetes and cardiovascular disease

ANYTHING ELSE TO KEEP IN MIND?

Used in combination with compounds like bicalutamide in prostate cancer
